Suspected cases of foot-and-mouth disease (FMD) have been reported in Hongseong, South Chungcheong Province and Sangju, North Gyeongsang Province.
Hongseong is the nation's largest livestock cluster while Sangju is the largest breeding place of domestic cattle, known as "hanwoo".
The Agriculture Ministry says it received the reports from hanwoo farms in the two areas.
Suspected cases have also been reported in Asan and Cheonan in South Chungcheong, Pyeongtaek in Gyeonggi Province and Goseong in Gangwon Province.
The latest reports come amid vaccinations administered on cattle nationwide. This is raising concerns that quarantine efforts may fail.
