Full factorial analysis of Mammalian and avian influenza polymerase subunits suggests a role of an efficient polymerase for virus adaptation.

Amongst all the internal gene segments (PB2. PB1, PA, NP, M and NS), the avian PB1 segment is the only one which was reassorted into the human H2N2 and H3N2 pandemic strains. This suggests that the reassortment of polymerase subunit genes between mammalian and avian influenza viruses might play roles for interspecies transmission. To test this hypothesis, we tested the compatibility between PB2, PB1, PA and NP derived from a H5N1 virus and a mammalian H1N1 virus. All 16 possible combinations of avian-mammalian chimeric viral ribonucleoproteins (vRNPs) were characterized. We showed that recombinant vRNPs with a mammalian PB2 and an avian PB1 had the strongest polymerase activities in human cells at all studied temperature. In addition, viruses with this specific PB2-PB1 combination could grow efficiently in cell cultures, especially at a high incubation temperature. These viruses were potent inducers of proinflammatory cytokines and chemokines in primary human macrophages and pneumocytes. Viruses with this specific PB2-PB1 combination were also found to be more capable to generate adaptive mutations under a new selection pressure. These results suggested that the viral polymerase activity might be relevant for the genesis of influenza viruses of human health concern.
Swine flu spreads to every Australian state, territory

Health authorities in every state and territory in Australia are now dealing with swine flu, after the Northern Territory reported its first case overnight.
A total of 255 people now have the virus.
The Northern Territory centre for disease control is monitoring the woman and a group of people she's been in contact with. They are being treated with tamiflu and are under home isolation.
The woman arrived on a flight from Melbourne to Darwin last Friday but has only now tested positive to the virus. It's unclear how many passengers she may have come into contact with but health authorities will try and track down them all.
It's the first case to be reported in the Northern Territory.
Most swine flu cases are in Victoria.
A total of 173 people are being treated there while in New South Wales there are 54 notifications.
Queensland has 15 cases, South Australia 6, there are four in the ACT and one each in Western Australia and Tasmania.

Scientists find more signs of bats’ role in spreading the deadly Ebola virus
Bats spread the Ebola virus to humans and play a pivotal role in disease outbreaks, evidence suggests.
Ebola, a filovirus, causes fever, vomiting, diarrhoea and sometimes bleeding. There is no treatment or vaccine and 25–90 per cent of infected people die. The virus is transmitted by direct contact with infected blood, body fluids and tissues.
The new findings have emerged from data collected in the remote Kasai-Occidental and Kasai provinces of the Democratic Republic of Congo (DRC) which experienced a large Ebola outbreak in 2007 in which 186 people died.

For the new study, researchers led by Eric Leroy from the International Centre for Medical Research in Franceville, Gabon, interviewed locals about the background of the Ebola cases. They were told that the annual migration of the fruit bat Hypsignathus monstrosus was particularly large in 2007.
Bats are an important source of protein in the area as wild animals are in short supply. They are often shot and then sold covered in blood.
The researchers believe the source of the 2007 outbreak was a man who bought bats at market. He survived, experiencing only a low fever, but his four-year-old daughter died after developing a sudden fever accompanied by vomiting.
A family friend who prepared the girl’s body for burial was subsequently infected and went on to infect 11 members of her family, all of whom died.
Researchers say their study suggests infection is only transmitted after prolonged contact with an infected person, meaning it may be easier to contain an outbreak than was previously believed.
Jean-Jacques Muyembe, epidemiologist at the DRC’s National Biomedical Research Institute and co-author of the study, told SciDev.Net that the team will continue to retrace events around outbreaks and carry out ecological studies in bats “to definitely prove that these bats are the direct vectors of Ebola to humans”.
Vital Mondonge, a co-author from the infectious disease section of DRC’s health ministry, says that the bat migration should be a focus of efforts to prevent Ebola infection.
Chimpanzees and gorillas are also known Ebola vectors. Bats, however, appear not to die from Ebola, suggesting they play a role in maintaining the virus in tropical forests.
Red Cross: Zimbabwe's Cholera Epidemic Expected To Infect 100,000th Victim Sometime This Week

Harare, Zimbabwa (NewsBahn) - Zimbabwe is experiencing its worst cholera outbreak in 15 years and it is likely that the 100,00th victim will develop the disease sometime this week, Red Cross officials say.
The International Federation of the Red Cross and Red Crescent Societies (IFRC) said in a statement Tuesday that there have been 98,309 cases officially reported, so far, with about 4,283 deaths.
Although the pace at which the number of new cases is being reported is beginning to slow, the Zimbabwe Red Cross Society (ZRCS) and the IFRC warned that the risk of infection remains.
"(The) eradication of cholera in Zimbabwe or the complete conclusion to this current epidemic is unlikely unless the underlying causes of the health crises are addressed," the joint ZRCS and IFRC report released Tuesday states.
"Central to this outbreak remains the almost complete collapse of Zimbabwe's basic water, sanitation and health infrastructure. Communities across the country are still without access to potable water and basic sanitation, and health facilities continue to be understaffed and under resourced," Red Cross officials said.
The next step is to make sure that households have clean water and the necessary sanitation facilities. However, Red Cross officials note that it will take years to rehabilitate Zimbabwe's infrastructure to accomplish that.
Cholera broke out 10 months ago. Widespread food insecurity made the situation worse.
Red Cross officials are trying to raise $3.5 million to repair existing water sources, develop more and build toilets. They are soliciting donations from the international community.

Dead cattle, fish, poultry and other animals floating in the water bodies in Atulia under Shyamnagar upazila bear witness to the destruction Aila caused. The living are in plight whereas the dead are lucky.
The air of Atulia is heavy with the cries of the marooned and stench of decomposed livestock. The road from Gumantoli Chuna Bridge to Noabeki Bazar reeks of death.
The need to pull out the decaying livestock from water is immense but no one sees the point as no high land was spared to bury them.
The river Kholpetua has flooded the entire union of Atulia; one of the worst Aila hit areas with tidal surge that wrecked the southwestern coastal belts of the country on May 25. As far as the eyes lead only water is visible as almost every village is reeling under knee to waist deep water.
Thousands of people of the area are still living on roads as not a single homestead was spared by the storm surge.
Almost all of the houses in these villages including Biralaxmi, Baduria, Uttar Atulia, Hawolbhangi, Chotokupor, Borokupor have been washed away. Almost every yard is now a pile of waste. Villagers are gathering whatever they can recover while many of them have abandoned the place.
The devastated and destitute families took refuge on the road, where some lost their valuables, most importantly, their precious cattle. Cattle means so much to these poor people of the country's southwestern part.
Scarcity of safe drinking water is now haunting the villagers.
With her daughter-in-law, octogenarian Gulaman Bibi of Biralaxmi village was seen in a long queue on the road for water. With a faint voice she said, “I do not want anything, just give me some water.”
Housewife Farida Khatun of Uttar Atulia was standing with her three-year-old daughter as she heard drinking water was on its way.
“My child has been suffering from diarrhoea since last night and is now dehydrated,” she said adding, “I need fresh water and saline to save her.”
“We do not have any pond left as most of them we used for drinking water have turned salty in the tidal surge,” she said adding that they cannot even cook since there is no high ground to place stoves or any fuel to light it up.
Masiron Bibi said, “I have lost everything. I have no house, clothes or medicine. We can go without food for few hours but no way we can survive without water.”
Many villagers, especially children, have got skin diseases because of the dirty and contaminated water used for bathing and other purposes.
Tube wells could not be installed here due to salinity and iron in the water, said Rabiul Islam, another resident of the village. So most of the villagers used to purify pond water for drinking, but these ponds have been inundated, he added.
Wading through waist-deep water, Jahidul came to collect water from the lone remaining tube well at Uttar Atulia village. “There is no tube well in three kilometres and the water from it is too inadequate,” he said adding that that tube well also went under water during the high tide.
Bimol Mallick of Baduria said he has to fetch water by boat, as his house is three kilometres away from Uttar Atulia.
Cooking with this salty water gives food an unpleasant taste and colour. Joyna Akhter said, “My children do not want to eat food cooked with the water as rice tastes salty and smells terrible.”
The locals are now resigned to the nightmare they are living through. They have no option but to crowd along the banks hoping for relief.

Veterinarians have warned the public not to transport, trade, or eat
livestock that have died as a result of haemorrhagic disease, as
outbreaks spread across the country. Some people in rural areas
continue to eat the meat of infected animals, as they are unaware of
the disease or uninformed about preventative measures.
The disease can easily spread to other livestock and people may fall
ill with diarrhoea if they eat animals that have died from the
disease, said Livestock and Fisheries Department veterinarian Dr
Phouth Inthavong. "We should incinerate and bury infected carcasses
at the place where the animals died, or at other places far from
where other livestock graze," he said. Infected livestock suffer from
a swollen stomach and neck and a fever, after eating grass or water
alongside other infected animals, he said.
People should bury dead livestock in pits that should be dug at least
1-1.50 meters [3-5 feet] deep to prevent dogs or pigs unearthing
infected carcasses. They should be covered with soil and then lime
sprinkled on the surface. These burial practices will prevent the
disease infecting other buffalo and cattle, he said.
Xaythany district Livestock and Fisheries Unit Head, Mr Thongtem
Luangxay, said illegal livestock movement around the country causes
the disease to spread. There have also been reported cases of black
leg disease in Xaythany district, Vientiane. This disease bears some
similarities to haemorrhagic disease but was less likely to spread
across the country, he said.
According to Dr Phouth, livestock with this disease suffer from
swollen legs and find it difficult to walk. But the disease is not
fatal if animals are treated appropriately and in a timely fashion.
Anthrax, which could potentially cause far greater damage to
livestock populations, was unlikely to occur in Laos at present,
despite outbreaks having occurred in past decades, Dr Phouth said.
"The most obvious symptoms humans infected with the disease display
are boils on their face or body. They will also become ill after
eating infected meat," he said.
In light of these risks traders must be responsible and consumers
should exercise caution. "We should not eat any dead animals infected
with any disease, both to prevent an outbreak from occurring and in
the interests of public health and safety," Dr Phouth said.

[According to the article, a hemorrhagic septicemia outbreak across
Laos has been reported and people who consume meat from dead animals
will get diarrhea. Nonetheless, the article has also reported an
outbreak of black leg disease in Xaythany district. The signs of the
disease reported from Xaythany are very similar to hemorrhagic
septicemia. It was also reported that people who consume dead animal
meat would have had a "boils on their face or body." Due to limited
details on the clinical signs and outbreak investigation data in both
animals and humans, a differential diagnosis of the disease as
anthrax, blackleg, and hemorrhagic septicemia is very difficult.

Since there are reports of diarrhea and "boils on their face or body"
after eating meat from dead cattle, it is worth mentioning that
anthrax in humans, especially its skin or cutaneous form, may begin
as a raised itchy bump that resembles an insect bite but within 1-2
days develops into a vesicle and then a painless ulcer, usually 1-3
cm in diameter, with a characteristic black necrotic (dying) area in

Intestinal anthrax is characterized by an acute inflammation of the
intestinal tract. Initial signs include nausea, loss of appetite,

initial symptoms may resemble a common cold. After several days, the
symptoms may progress to severe breathing problems and shock. (more

Egypt reports 77th bird flu case
CAIRO, May 30 (KUNA) -- The Egyptian Health Ministry confirmed on Saturday the 77th infection case with H5N1 virus.
A female baby, aged one year and two months, contracted the highly pathogenic virus because she got in contact with domestic birds in a village located in Daqahliya, north Cairo, the ministry's spokesman Dr. Abdulrahman Shahin said in a press briefing.
The baby was hospitalized five days ago, suffering from bird flu-like symptoms including high temperature, running nose, and respiratory disorder.
She was diagnosed as H5N1 positive so she has been treated with the antiviral drug Tamiflu, Dr. Shahin revealed.
The victim is being relocated to Manshiyat Al-Bakri Hospital, Cairo, for further medication, he added.
The case pushed to 77 Egypt's national tally of the fatal disease with the death toll amounting to 27 since the outbreak of the highly contagious virus in February, 2006.
The National Higher Commission against Bird Flu has adopted a new preventive methodology to control the spread of the H5N1, a subtype of the influenza A virus commonly-known as bird flu which can cause illness in humans. The methodology involves the medical and municipal authorities as well as the mass media. (end)

The epidemiologist was talking about the city’s response to the outbreak at the New York Academy of Sciences, when he let slip the fact that his 9-year-old daughter was ill with swine flu. Scientific American editor Christine Soares was seated in the audience and says “nobody blinked” as Harper casually recounted his all-too-personal experience with the virus.
Swine flu has killed four people in New York State, and hospitalized more than 100, including 10 patients currently on ventilators in the city. The outbreak seemed to be waning in recent weeks, but yesterday the city closed 6 additional schools, bringing the total to 13. Harper has himself been popping up in the news media urging parents with sick children to stay home from school – and he’s apparently heeding his own advice.
“The big picture is not the number of deaths in any given city,” Harper told Newsday on Wednesday, “The big picture is how many cases are coming.”
